Kumulus hücrelerinin in-vitro olarak oosit maturasyonunu ne ölçüde etkilediğini araştırmak amacıyla GATA IVF merkezine farklı zamanlarda başvuran 40 hastadan follikül aspirasyonu yapılarak toplanan 80 immatür (PI), 100 intermediate (MI) olmak üzere toplam 180 insan oositi incelendi. Oositler rastgele iki gruba ayrıldı. İlk grubun kumulus ooforusları temizlendi (kumulussuz), 2. grup ise kumulusları temizlenmeksizin (kumuluslu) çalışıldı. İn vitro maturasyon süreçleri 4er saat arayla 36. saate kadar gözlendi. Farklı sürelerde maturasyonunu tamamlayarak MIIye ulaşan kumuluslu ve kumulussuz oositler belirlendi. IVMa ulaşan kumulussuz oosit oranı %24.4 (22/90) olduğu halde, kumuluslu olanlarda bu oran %60 (54/90) bulundu (t=5.159, p<0.01). En yüksek IVM oranına 12-16. saatler arasında (%17.7) ve kumuluslu olarak inkübe edilen oositlerde rastlandı. Bu bulgular ışığında, kumulus hücrelerinin varlığının IVMda çok önemli bir rol oynadığı söylenebilir.
The objective of this study was to investigate the effects of cumulus oophorus cells on in vitro oocyte maturation. A total of 180 oocytes (80 immature - prophase I and 100 intermediate - metaphase I) were collected from 40 patients. These oocytes were randomly distributed into 2 groups. The first group oocytes were denuded and seperated from cumulus oophorus cells (cumulus free). No denudation was done in the second group. During the incubation period the stage of the in vitro maturation of the cumulus free and intact oocytes were observed every 4 hours up to 36 hours. The undenuded (cumulus intact) oocytes in the first group, were found to reach metaphase II stage 60% (54/90) whereas, denuded oocytes reached 24% (22/90). The peak for reaching metaphase II stage were observed in the undenuded group of oocytes at 12-16 hours. It is obvious that the presence cumulus oophorus cells play a very important role for in-vitro maturation.
