Cumulus granulosa cells of poor, normal and over responder IVF patients have disparities in organization of cytoplasmic organelles and actin filaments: a confocal microscopic study

OBJECTIVE: The aim of this study is to characterize the cytoplasmic organelle and actin organization of human cumulus cells obtained from IVF cycles performed in poor, normal and over responder patients by fluorescence confocal imaging. DESIGN: Three groups of patients were selected (poor, normal and over responders) regarding the number of oocytes obtained.Cumulus cells were stained with specific markers and observed under confocal microscope to reveal the differences. MATERIALS AND METHODS: Cumulus cells from three patient groups (n¼20/each) were isolated with respect to their proximity to zona pellucida. Cell suspensions were fixed in acetone and 10% formalin.Cells were stained with hematoxylin/eosin,anti-mitochondria,anti-transgolgi,anti-endoplasmic reticulum antibodies and phalloidin.Images were taken under Eclipse 90i Nikon confocal microscope. RESULTS: H&E stainings showed in all groups granulosa cells had little cytoplasm with abundant granules and centrally localized nuclei.The cytoplasm was poor in actin filaments with the interruption of filamentous structure by surrounding organelles.The periphery of the cells were demarcated by actin decoration more prominently in outer cumulus cells of poor responders.Golgi bodies were close to nucleus at one side as a crescent rim with no specific alterations regarding location or the patient groups. Mitochondria were diffusely distributed throughout the cytoplasm in inner and outer cumulus cells,but accumulation of mitochondria were more abundant and organized in normal responders than poor or overresponders.The endoplasmic reticulum was finely allocated throughout the cytoplasm with prominent cisternae near to nucleus.This distribution was not conspicuous in over responders and less evident in poor responders.Demonstrative pictures were taken to show the specific differences. CONCLUSION: Confocal images revealed cumulus cells of poor, normal and over responder patients had differences in the distribution and intensity of major organelles and actin in various aspects.
