Differential distribution of bFGF and bFGF receptors in human cumulus granulosa cells of poor, normal and over responder IVF patients

OBJECTIVE: The aim of this study is to discover the discriminations in distribution of bFGF and bFGF receptors in human cumulus cells obtained from IVF cycles performed in poor, normal and over responder patients. DESIGN: Three groups of patients were selected (poor, normal and over responders) regarding the number of oocytes obtained.Cumulus cells were stained with specific markers and observed under confocal microscope to reveal the differences. MATERIALS AND METHODS: Cumulus cells obtained from three patient groups (poor, normal and over responders, n¼20/each group) were isolated with respect to their proximity to zona pellucida, as cells at the outer most area of the follicle were isolated mechanically, while the ones neighboring the zona pellucida were isolated enzymatically.The cell suspensions were fixed in absolute acetone at -20C for 10 minutes. Cells were stained with anti-bFGF and anti-bFGF receptor.Confocal microscope images were taken under Eclipse 90i Nikon confocal microscope. RESULTS: Preliminary results showed that bFGF staining was not observed in neither cell cytoplasm nor the extracellular matrix area in all groups. However its receptors were well defined inclusively in the certain cellular membrane areas extending into cytoplasm. Although all groups showed similar staining in both outer and inner cumulus cells,the pattern was most prominent in outer cumulus cells of over responder patients. Demonstrative pictures were taken to show the specific differences. CONCLUSION: Since bFGF is known to be located in the cumulus cells and oocytes of primordial and primary follicles (to influence the development and the transition of primordial follicle), non-staining of bFGF in cumulus cells of induced patients can be expected. However the presence of bFGF receptors is important since this may demonstrate that bFGF can work through its receptors for its many activities (as antiapoptotic or mitogenic activity, etc.) if it is provided in the microenvironment of induced follicles.
